Two Common Over-the-Counter Compounds Reduce COVID-19 Virus Replication by 99% in Early TestingA pair of over-the-counter compounds has been found in preliminary tests to inhibit the virus that causes COVID-19, University of Florida Health researchers have found. The combination includes diphenhydramine, an antihistamine used for allergy symptoms. When paired with lactoferrin, a protein found in cow and human milk, the compounds were found to hinder the SARS-CoV-2 virus during tests in monkey cells and human lung cells. The findings by David A. Ostrov, Ph.D., an immunologist and associate professor in the UF College of Medicine’s department of pathology, immunology and laboratory medicine and his colleagues, are published in the journal Pathogens. “We found out why certain drugs are active against the virus that causes COVID-19. Then, we found an antiviral combination that can be effective, economical, and has a long history of safety,” Ostrov said. Due to his earlier research with colleagues at UF, Ostrov already knew diphenhydramine was potentially effective against the SARS-CoV-2 virus. The latest discovery has its roots in a routine meeting of scientists with the Global Virus Network’s COVID-19 task force. One researcher presented unpublished data on federally approved compounds that inhibit SARS-CoV-2 activity, including lactoferrin. Like diphenhydramine, lactoferrin is available without a prescription. Ostrov thought about pairing it with diphenhydramine and ran with the idea. In lab tests on human and monkey cells, the combination was particularly potent: Individually, the two compounds each inhibited SARS-CoV-2 virus replication by about 30%. Together, they reduced virus replication by 99%. |

(carbon20 What is wrong with you ???) https://en.wikipedia.org/wiki/Fluticasone_propionate or https://en.wikipedia.org/wiki/Fluticasone_propionate is protecting against (severe CoViD0-a Belgian study shows. Could not find a link in English so google translate had a job; DJ There may be lots of effective medications limiting risks (either getting infected or limiting infection) but we may need to link medication with CoViD (and the correct group of patients). |

Highly Specific Sigma Receptor Ligands Exhibit Anti-Viral Properties in SARS-CoV-2 Infected Cells1 Department of Pathology, Immunology and Laboratory Medicine, University of Florida College of Medicine, Gainesville, FL 32610, USA 2 Department of Geography, University of Florida College of Liberal Arts and Sciences, Gainesville, FL 32611, USA 3 Emerging Pathogens Institute, University of Florida, Gainesville, FL 32610, USA 4 Department of Biochemistry, Microbiology, and Immunology, University of Saskatchewan, Saskatoon, SK S7N 5E5, Canada 5 Division of Oncology, College of Medicine, University of Saskatchewan, Saskatoon, SK S7N 5E5, Canada 6 Vaccine and Infectious Disease Organization-International Vaccine Centre, University of Saskatchewan, Saskatoon, SK S7N 5E3, Canada 7 Cancer Research Department, Saskatchewan Cancer Agency, Saskatoon, SK S7N 5E5, Canada 8 Department of Medicinal Chemistry, University of Florida College of Pharmacy, Gainesville, FL 32610, USA 9 Department of Pharmaceutics, University of Florida College of Pharmacy, Gainesville, FL 32610, USA 10 Translational Drug Development Core, University of Florida Clinical and Translational Sciences Institute, Gainesville, FL 32610, USA * Author to whom correspondence should be addressed. Academic Editor: Lawrence S. Young Pathogens 2021, 10(11), 1514; https://doi.org/10.3390/pathogens10111514 Received: 14 October 2021 / Revised: 10 November 2021 / Accepted: 17 November 2021 / Published: 20 November 2021 (1) Background: There is a strong need for prevention and treatment strategies for COVID-19 that are not impacted by SARS-CoV-2 mutations emerging in variants of concern. After virus infection, host ER resident sigma receptors form direct interactions with non-structural SARS-CoV-2 proteins present in the replication complex. (2) Methods: In this work, highly specific sigma receptor ligands were investigated for their ability to inhibit both SARS-CoV-2 genome replication and virus induced cellular toxicity. This study found antiviral activity associated with agonism of the sigma-1 receptor (e.g., SA4503), ligation of the sigma-2 receptor (e.g., CM398), and a combination of the two pathways (e.g., AZ66). (3) Results: Intermolecular contacts between these ligands and sigma receptors were identified by structural modeling. (4) Conclusions: Sigma receptor ligands and drugs with off-target sigma receptor binding characteristics were effective at inhibiting SARS-CoV-2 infection in primate and human cells, representing a potential therapeutic avenue for COVID-19 prevention and treatment. I still say vaccines are the way out ,not snake oil remedies..... Seems ai ,who is totally against vaccines ,will jump onto any ,sudoscience, To justify not taking a vaccine, governments all over the world are imploring people to take, Why would I listen to or read something from someone who is so anti Vax.......?? Or from a university that's under investigation for fraud ?..... Nah I'm no clown...... Take care all π·ππ |

Good find, thank you AI :-) Antihistamine would 'calm' the immune system so that actually makes sense. My daughter has celiac and say she accidentally eats wheat. If we are quick enough with antihistamine her symptoms are greatly reduced. Obviously I wouldn't use it alone lol but I think it might help. |

I (who am double-vaxxed and due in Jan/Feb to be boosted) think that if I were to contract Covid, I would want all available tools to be utilized to lessen its impact/save my life, or address whatever condition I was in. Here are some studies going on into repurposing some common meds for off-label use to address Covid: https://www.ncbi.nlm.nih.gov/pmc/articles/PMC7713548/ https://www.ncbi.nlm.nih.gov/pmc/articles/PMC8354566/ If the science supports the safety/effectiveness, and I trusted the doctor who was treating me, I would take any of these treatments. Diphenhydramine and Fluticasone are already a part of my toolkit for allergies/asthma, and it may interest some that Fluticasone has been used to treat some forms of lymphoma. |

Thank you! I take Fluticasone for asthma and allergies (both inhaled oral and sinus) and I take diphenhydramine every 4 hours for allergies. I will take every tiny possible advantage I can get! |

Imagine if this study leads to a 99% effective, economical preventive and treatment of covid which has the added benefit of a long history of the components being safe. Think Sudafed type like for a cold. Covid would be knocked out in short order. I think it's a very positive thing and a promising study and bears watching further developments. |

I had a conversation a few years ago with our GP who told me that many medicines had well known secondary uses. She was prescribing a cream for a rash one of my children had and told me that this cream was the best on the market for burns. So now I make a point of asking what the alternative uses for them are. I've kept all our old medications, just in case. |

ViQueen thank you for links. Very interesting... And I agree with others that most medicines can have many uses :-) *I am double vaccinated too but have never been one to rely on something fully. |
