Potential Human Adaptation Mutation of Influenza A(H5N1) Virus, Canada

Emerg Infect Dis vol. 20 (9) - September 20146 August, 2014

In December 2013, influenza associated with pandemic influenza A/H5N1 was reported in Canada in a patient who had traveled to China; the patient died in January 2014. In the absence of direct poultry contact by the patient, the possible route of transmission and infection, often influenced by receptor-binding properties of the virus, requires special attention. The investigators note two previously reported mutations, R189K and G221R, in the hemagglutinin protein in the virus isolated from the patient. They write that both mutations are found in the immediate receptor-binding pocket, and G225R has been known to change specificity of an H3N2 virus toward human erythrocytes. The authors note that the two receptor-binding pocket mutations were not seen in the most closely related Asian H5N1 sequences.
The researchers from Singapore and Canada suggested that G225R could incur a relative predicted increase in binding to the human-like receptors, the role of R193K is less clear with a slight predicted tendency of favoring avian-like receptors.

Every time we write H5N1 off, it pops back up with something new. I've yet to see anything that changes my belief that influenza is the virus that could potentially bring us to our knees. It mutates at a hellacious rate, reassorts with ease, spreads like wildfire, and can kill as efficiently as just about anything out there. No other pathogen has as many pandemic cards in it's deck, and nothing else has been handed the opportunities that influenza has in the form of wet markets and factory farming.

Maybe, maybe not, Kilt. This bad boy is certainly taking it's sweet time. But even if it doesn't manage the last few mutations it needs, there are numerous strains brewing in the wild and domestic poultry that could jump without warning.

Hey guys, there is going to be a Pandemic....a huge one and it does not matter what it is it is coming. Why is because there are too many people.

In the old days it was because of close dirty quarters, no containment of rodents, no meds, and war where men were in close quarters.

I am upping my preps slow but sure. By mid October I will be ready as I can be for what comes!

Couldn't agree more, FluMom. Overpopulation is a huge factor in the propagation and spread of disease.

there are 2 kinds of mutation - drift and shift

this bug is playing Russian Roulette with its RNA and the chamber is loaded.

The canadian woman had a new mutated line where its almost a pandemic strain - this is very concerning and very close.

All it takes is for someone to catch H5N1 and a Human flu H3N2 for example and a recombined menace is made and will sweep the world.

Believe me Kilt, I went over it quite a few times when you first posted it. And I agree - it's significant in it's implication that maybe H5N1 is not as genetically stable as we'd begun to think. A lot of us had written it off as a pandemic threat, but I guess that might be a little naive given flu's innate ability to mutate. And maybe it won't be the next pandemic strain, but it may bring something to the viral mix and match party that spawns the next big one.
