Researchers seeking H7N9 influenza vaccine uncover immune system camouflage

Researchers working to develop an avian influenza A (H7N9) vaccine have discovered the virus uses a kind of camouflage to avoid the immune system and to evade positive results in vaccination.

H7N9 caused a serious outbreak in China in 2013. Since that time, scientists have been concerned about its high rate of lethality as well as its pandemic potential, making developing an H7N9 vaccine a top priority.

Unfortunately, none of the vaccine candidates has produced strong immune responses to counteract the virus. Researchers have discovered this is because the virus uses immune camouflage to remain undetected.

"The original observation of low H7N9 T-cell epitope content was made before any data were available on vaccine efficacy," Anne De Groot, EpiVax Inc. CEO and senior author and director of the Institute for Immunology and Informatics at the University of Rhode Island, said. "It turns out that we were absolutely correct. By comparison with H1N1 and H3N1, H7N9 vaccines are far less immunogenic."

This discovery could change the approach to developing vaccines for other diseases as well.

"It appears that this new mechanism of immune escape may be common to quite a few human pathogens," De Groot said. "We are working on validating several other peptides, some from common seasonal strains of influenza and some from pathogens like HIV and M. tuberculosis that do live for a long time inside their hosts."

Arirish, H5 and H7 experimental vaccines have proved poorly immunogenic in the past – requiring unusually large amounts of antigen (up to 12x normal), spread across two shots several weeks apart  - to induce a good immune response.

One way around this obstacle is to add an adjuvant -  proprietary ingredients that are added to vaccines to increase the recipient’s immune response and that can dramatically lower the amount of antigen needed in a vaccine.

An announcement from the NIH: H7N9 Vaccine Candidate Works Much Better With An Adjuvant.  According to their study, which investigated an adjuvant called  MF59.

The bottom line, without an adjuvant, only 2% of vaccine recipients saw an acceptable immune response (HAI Titer > 40). Two 15 µg shots were still required, but those who received ASO3-adjuvanted vaccines were more likely to mount an acceptable immune response (84%) compared to those who received the MF59 adjuvanted formula (57%).

Their is a lot of obfuscation out there but your seem to be covering the right stuff, you strike me as a person that listens more than they speak and it's a pleasure reading your posts.

Thanks Guests! Like everyone else here I have my opinions but that's all they are! I found AFT after my mother passed in 09 and I started coming here to learn about influenza and to share what I find on the net. Arguing the yeas and nays of vaccines or whether the Pope reads the bible seems a great waste of time to me but then some have more time on their hands I suppose! keep up the search!
